NanoConjugates
Background

OurNanoConjugates were evolved in a multi-level in house selection process starting with cDNA libraries from immunized alpaka PBMC cells or a random single stranded synthetic DNA library coding for the variable domain of heavy chain only antibodies. The processing included multiple rounds of in vitro selection or more precisely phage and ribosome display for enrichment of putative binders to the adressed target antibodies and Next Generation Sequencing as well as bioinformatical analysis of enrichment patterns. Promising candidate pools were screened for their ability to bind human serum antibodies in an diagnostic ELISA assay and the best performing ones were further characterised by protein expression analysis. As reporter module, we linked a discrete protein module with either acridinium or horseradish peroxidase. The final constructs were validated regarding their applicability in IVD platforms, such as LineAssay or bead based automated CLIA.
